Following this, VB12 conjugated to biotin competes with the antigen in the sample to bind the capture antibody or antigen. Unbound conjugate and sample antigen is washed away and bound conjugate is then reacted with avidin conjugated to horseradish peroxidase. TMB substrate is used for color development at 450nm. Color development is inverse proportional to the concentration of active VB12 in the samples. A standard calibration curve is prepared along with the samples to be measured using dilutions of VB12.
